Epitalon (also known as Epitalon and Epitalamin) is a short synthetic peptide known for its ability to activate the enzyme telomerase and stimulate melatonin release. First developed in Russia in the 1980s, Epitalon has demonstrated an ability to slow age-related changes in the reproductive and immune systems and to increase lifespan in mice and rats. Although it is of primary interest to researchers in anti-ageing, Epitalon has shown significant effects in certain types of cancer, infectious diseases and DNA regulation.
Epitalon is of interest in anti-ageing medicine because of its potential benefits:
Telomeres: Epitalon may increase the length of telomeres, the protective caps at the ends of chromosomes that shorten with age. This may slow cellular ageing.
Cell repair: The peptide may stimulate cell renewal, which is important for tissue health.

As stated above, Epitalon has a positive effect on the gene that regulates MMP2. MMP2 is a protein found in connective tissue such as skin. Studies in rodents show that Epitalon not only activates this gene, it also activates fibroblasts, the cells that produce and maintain MMP2, as well as other components of the extracellular matrix such as collagen and elastin. Mice exposed to Epitalon show a 30-45% increase in fibroblast activation. By activating fibroblasts, Epitalon may help accelerate healing processes and compensate for the natural decline in skin structure and integrity that occurs with ageing. Further evidence of Epitalon's benefits for the skin is that it reduces caspase-3 activity. Caspase-3 is an enzyme in the pathway of apoptosis, or programmed cell death. By reducing caspase-3 activity, Epitalon helps protect fibroblasts and other skin cells, keeping them alive and healthy for longer periods.
Daily administration of Epitalon to rats with cancer was found to reduce tumour growth. The peptide not only reduces tumour growth but also prevents these tumours from metastasising to distant tissues. Epitalon is currently being studied as a potential treatment for HER2/neu-positive breast cancers and is of interest in preventing the development of certain types of leukaemia and testicular cancer. There is some evidence that Epitalon activates the gene for the PER1 protein, which is found in the hypothalamus. PER1, which helps regulate circadian rhythms, is underexpressed in patients with cancer. It is unclear whether this underexpression precedes the development of cancer and therefore contributes to cancer growth, or is a consequence of cancer development. It is clear that the protein affects cancer growth once cancer is established. Controlling PER1 expression may be one way to naturally slow tumour growth. Studies show that PER1 expression sensitises cells to the effects of radiation and may therefore help reduce the radiation doses needed to treat certain cancers, which would reduce immediate side effects and the occurrence of secondary tumours after high doses of radiation.
Melatonin, which is associated with sleep and ageing, is produced by the pineal gland. Studies in rats show that Epitalon and similar peptides affect both melatonin synthesis and release by influencing the expression of two proteins (arylalkylamine-N-acetyltransferase (AANAT) and the pCREB transcription protein)[15]. Both genes play an important role in melatonin production and the circadian (day/night) control of its release. Studies in monkeys indicate that Epitalon restores normal melatonin release [16].
A trial in rats suffering from retinitis pigmentosa showed that Epitalon improved outcomes in 90% of patients. The peptide appears to help preserve the normal structure of the eye while enhancing the bioelectrical function of the retina needed for vision. Epitalon has minimal side effects, low oral bioavailability and excellent subcutaneous bioavailability in mice. The dosage per kilogram of body weight in mice is not proportional to that in humans. Use:
Correct use and dosing of Epitalon depend on the patient's specific needs and goals. It is usually prescribed at 3 mg to 5 mg once daily, and the dose may be increased to 10 mg.
Additional instructions for use:
Set the pen dial by turning it from 18 to 30 (1 unit equals 1 increment on the dial) and inject into the abdomen or front of the thigh.
After injecting the product, wait 10 seconds before removing the needle. Rotating the injection site is recommended.
A cycle with this peptide lasts 15-20 days, followed by a break of about 15 days.
